-ENLACE Research: Molecular Cloning of Human Lactate Dehydrogenase Protein Coding
Sequence in E. Coli (June-July 2023, September-October 2023):

  • Reverse transcription of HepG2 mRNA and PCR Reaction
  • Usage of Argorose Electrophresis gel to (post PCR Reaction) to amplify kb strand (length of
    literal DNA protein using specific primers (cDNA and hLDH)
  • Use centrifuge machinery (seperate proteins, lipids, etc…)
  • Restriction Digest (both NcOI and HindIII) of the putative ~1kb hLDH amplicon representing the hLDH ORF and the PSE420 expression vector: Cut (lanes A8, B7; ~4.3 kb); Uncut (lanes
    A9, B3, B8; ~3.5kb).
  • Performed another ligation reaction followed by transformation into DH5-alpha competent cells
  • PCR-screen of colonies using insert specific primers, using ampicillin resistant colonies as a source of template and subsequent agarose gel electrophoresis.
  • Restriction-digest and PCR screen of isolated plasmids. Colonies screened with PCR were grown overnight and their plasmids subjected to single and double digest with HindIII and NCOI-HindIII, respectively (lanes 1-6).

  • Dealt with Isochrysis Galbana algae to try and harvest/ cultivate and produce the algae
    continuously

  • Use of pH meters, water tank heating systems, as well as nutrient-sufficient food for algae

  • Devised a poster board project pertaining to Bioretentions and the concepts/practical purposes they serve on communities.
  • Looked at data regarding water loss and how impervious (man-made structures) deter water away rather than being naturally absorbed and seeped into the soil.
  • Performed research and looked at real analytical data from Bioretention zones all across the San Gabriel Valley (local)
  • Sought data and science-based research online (factual data) that pertains to the benefits of having Bioretention zones and their water-collecting tactics that can supply neighborhoods and communities through various filtration systems and other scientific methods.
