Accomplishments

  • Assessing Compound Self-aggregation in Drug Discovery and Development
  • In aqueous solution small molecules exist in a unique three-phase equilibrium between single lone- tumbling molecules, self-associated aggregates, and solid forms
  • The nature of the equilibria profoundly dictates compound properties; however, characterization of this phenomenon remains relatively unexplored, particularly from a pharmaceutical perspective
  • For instance, employing dynamic light scattering (DLS) has determined that some molecules can self-assemble into micelle- like colloids and bind proteins, resulting in false-positive results in drug screens
  • However, the existence of smaller aggregates or nano-entities have historically been undetectable and thus their corresponding properties remain uncharacterized
  • Here, we present methodologies for monitoring the molecular properties of compound aggregates
  • First, we employ nuclear magnetic resonance (NMR) spectroscopy as a practical and valuable tool to directly observe lone molecules, small nano- entities, and indirectly, by observing large micelle-like aggregates
  • In this study, we also employ

  • This method clearly exposes large micelle-like particles of variable size
  • We employ the long-held assumption that molecular aggregates cannot form within cells
  • We discovered that dyes and drugs can form aggregates intracellularly in HeLa and Huh-7 cells, having discrete nuclear or cytoplasmic localization
  • As determined by confocal laser scanning microscopy (CLSM)
  • Implementing the above strategies expose a wide variety of molecular compound behaviors in solution and within cells
  • Given this new set of tools, screens can be implemented to aid in the understanding of the physical properties of compound aggregates such as, toxicity, off-target activity and loss of efficacy
  • These molecular properties provide new insights on how molecules behave, in vitro and in vivo, which can be exploited for drug new properties such as encapsulation and delivery systems
